Diopter (d): A term used to identify the refractive (light bending) capacity of a lens. In magnifiers, there is a direct correlation between focal length and diopter. To find the diopter of a magnification lens, follow these steps. With the eyes 10″ above the lens, move the object to be viewed to the point the greatest distance below the lens where it remains in sharp focus.

Measure this distance and divide into 1 meter (39.37″). The result is the diopter of the lens – e.g., if the object is at a 13″ distance than it is a 3-diopter lens (39.37/13 = 3d). Each diopter increases the size of the viewed object by 1/4 (25%) when the object is at its full focal length from the lens.

Field of View: The distance across the lens surface to which the viewer brings both his eyes (note: eyes should be 10″ above the lens). It is important to note that as magnification increases, meaning the lenses used are stronger, viewing areas and focal length decrease.

Magnification: The degree to which the viewed object is enlarged. Magnification is usually expressed by a number followed by an “x”, the symbol used to express power or the size of the object in relationship to its actual size.

Diopter indicates the amount of curvature of a lens. The thicker the lens, the bigger the curvature.
To find the approximate magnification level of a lens, divide its diopter by 4, and add 1.
The equation is:
Magnification = Diopter ÷ 4 + 1

Diopter and Magnification for example:
For a 3-diopter lens of a magnifier, its magnification = ¾ + 1 or 0.75 + 1 = 1.75x. Objects viewed under a 3 diopter lens will appear 1.75X larger than normal.
For a 5-diopter lens of a magnifier, its magnification = 5/4 + 1 or 1.25 + 1 = 2.25x. Objects viewed under a 5 diopter lens will appear 2.25X larger than normal.
For a 8-diopter lens of a magnifier, its magnification = 8/4 + 1 or 2 + 1 = 3x. Objects viewed under a 8 diopter lens will appear 3X larger than normal.

Microscope accessories and spare parts function

There are two optical systems in a compound microscope: Eyepiece Lenses and Objective Lenses:

1.1  Eyepiece or Ocular
is what you look through at the top of the microscope. Typically, standard eyepieces have a magnifying power of 10x. Optional eyepieces of varying powers are available, typically from 5x-30x.
Eyepiece Tube holds the eyepieces in place above the objective lens.
Binocular microscope heads typically incorporate a diopter adjustment ring that allows for the possible inconsistencies of our eyesight in one or both eyes. The monocular (single eye usage) microscope does not need a diopter. Binocular microscopes also swivel (Interpupillary Adjustment) to allow for different distances between the eyes of different individuals.

1.2 Objective Lenses
are the primary optical lenses on a microscope. They range from 4x-100x and typically, include, three, four or five on lens on most microscopes. Objectives can be forward or rear-facing.Nosepiece houses the objectives. The objectives are exposed and are mounted on a rotating turret so that different objectives can be conveniently selected. Standard objectives include 4x, 10x, 40x and 100x although different power objectives are available.
1.3 Coarse and Fine Focus knobs
are used to focus the microscope. Increasingly, they are coaxial knobs – that is to say they are built on the same axis with the fine focus knob on the outside. Coaxial focus knobs are more convenient since the viewer does not have to grope for a different knob.
1.4  Stage
is where the specimen to be viewed is placed. A mechanical stage is used when working at higher magnifications where delicate movements of the specimen slide are required.
1.5   Stage Clips
are used when there is no mechanical stage. The viewer is required to move the slide manually to view different sections of the specimen.this is stand base of microscope accessories.

1.6 Aperture is the hole in the stage through which the base (transmitted) light reaches the stage.

1.7  Illuminator is the light source for a microscope, typically located in the base of the microscope. Most light microscopes use low voltage, halogen bulbs with continuous variable lighting control located within the base.

1.8 Condenser is used to collect and focus the light from the illuminator on to the specimen. It is located under the stage often in conjunction with an iris diaphragm.

Iris Diaphragm controls the amount of light reaching the specimen. It is located above the condenser and below the stage. Most high quality microscope include an Abbe condenser with an iris diaphragm. Combined, they control both the focus and quantity of light applied to the specimen.

Condenser Focus Knob moves the condenser up or down to control the lighting focus on the specimen.

Difference between digital microscope and stereo microscope
1.1  difference between digital microscope and stereo microscope  is the magnification. With a stereo microscope the magnification is found by multiplying the lens magnification by the eyepiece magnification.

1.2  Since the digital microscope does not have an eyepiece. Instead the magnification for a digital microscope is found by how many times larger the sample is reproduced on the monitor. Therefore, the magnification will depend on the size of the monitor.

1.3 The average digital microscope system, which has a 15″ monitor, would result in an average difference in magnification between an optical microscope and a digital microscope of about 40%. Thus the magnification number of a stereo microscope is usually 40% less than the magnification number of a digital microscope.

Since the digital microscope has the image projected directly on to the CCD camera, it is possible to have higher quality recorded images than with a stereo microscope.
